You have two separate but joined systems on water. One is to use when hooked up to a water line. For that It is safer to have a pressure regulator as some systems near mountains may have extremely high pressure. Normally any reg set to less than 50 will be fine. I keep the cheap preset ones and don't worry what to set.

On city water, turn the valve to normal when just using the water on hose and fill when you want it to fill the tank. No help needed from the pump so leave it off but do remember to turn the knob back to normal after filling the tank or you will get very low flow at faucets.
The only time you need the pump on is after you have water in the tank, want to use some but are not hooked to city water. Turn the pump on and you likely will hear it pump until it gets pressure up on all the pipes and then stop. When using water the pump will cycle on and off to keep the pressure up and water flowing. We turn the pump off after we are done but it can be left on if you feel like it.
